Marfan syndrome is a hereditary connective tissue disorder that affects many parts of the body, including the heart, eyes, blood vessels, and skeleton. Marfan syndrome is inherited in an autosomal dominant pattern. This means that a person only needs to inherit the defective gene from one parent to be affected. However, in some cases, individuals may develop Marfan syndrome due to a spontaneous mutation in the FBN1 gene, which encodes the protein fibrillin-1. These spontaneous mutations are a risk factor for those without a family history of the disorder.

Family history is one of the most significant risk factors. A detailed family medical history can reveal whether there are relatives with Marfan syndrome or related connective tissue disorders. Such history prompts healthcare providers to undertake genetic testing and regular screenings to detect early signs of the condition, which can significantly improve outcomes.

Genetic mutations are central to the development of Marfan syndrome, but specific mutations within the FBN1 gene may influence the severity of the disease. Some mutations result in milder symptoms, while others lead to more severe cardiovascular or skeletal issues. Understanding these genetic variations can help predict disease progression and tailor management strategies.

Although the primary risk factor is genetic, certain features can alert clinicians to the possibility of Marfan syndrome. For example, individuals with a family history of the disorder, or those exhibiting characteristic physical features, such as tall stature, long limbs, arachnodactyly (long fingers and toes), scoliosis, or chest wall deformities, are more likely to carry the genetic mutation. Early diagnosis through physical examination and genetic testing can mitigate the risk of life-threatening complications, especially related to the heart and blood vessels.

It is also worth noting that although Marfan syndrome is not caused by lifestyle or environmental factors, individuals with the condition need to be cautious. High-impact activities or strenuous sports might increase the risk of aortic dissection or rupture, which are serious complications. Therefore, risk management involves not only genetic understanding but also lifestyle modifications and regular medical monitoring.
